PROPERTIES

Negazole is a synthetic 5-nitroimidazole derivative with antiprotozoal and antibacterial activity. Negazole is well absorbed after oral administration and its bioavailability approaches 100%. It appears in most body tissues and fluids and achieves concentrations similar to those in plasma. The majority of a dose of Negazole is excreted in the urine. Negazole is active against many organisms including Trichomonas vaginalis, Entamoeba histolytica, Bacteroides sp., Clostridium sp., Fusobacterium sp., Giardia lamblia, Peptococcus and Peptostreptococcus sp.

INDICATIONS

  1. Trichomoniasis, non-specific vaginitis.
  2. Intestinal and hepatic: amoebiasis.
  3. Giardiasis.
  4. Vincent’s infection (acute ulcerative gingivitis).
  5. Anaerobic bacterial infections.
  6. Negazole is used to eradicate H.pylori in peptic ulcer patients with Other products.

DOSAGE AND ADMINISTRATION

To be given orally.

Negazole dosage for trichomoniasis

  • Adults : 2 g of Negazole. given either as a single dose or in 2 divided doses, or 250 mg three times daily for 7 consecutive days. If pregnant women treated during 2nd, 3rd trimester. the one-day course of therapy should not be used. Spouse should be treated concurrently.
  • Children: 5 mg/kg  body weight three times daily for 7 days.

    • Negazole dosage for Non-specific vaginitis

    500 mg twice daily for 7 days. Spouse Should treated concurrently.

    Negazole dosage forIntestinal and hepatic amoebiasis

  • Adults : 500-750 mg three times daily for 5-10 days.
  • Children: 35-50 mg/kg body weight/day divided into 3 doses for 10 days.

    • Negazole dosage for Giardiasis

  • Adults: 2 g once a day for 3 days; or 250 mg three times a day for 5 to 7 days.
  • Children: 15 mg/kg body weight/day divided into 3 for 5 to 7 days.

    • Negazole dosage for Anaerobic bacterial infections 

  • Adults : 500 mg every 8 hours.
  • Children: 20-30 body weight/day divided into 3-4 doses.

    • Negazole dosage for eradication of H.pylori

    250 mg four times a day in combination with other products.

    CONTRAINDICATIONS

    1. Hypersensitivity to Metronidazole.
    2. During the first trimester of pregnancy.

    WARNINGS

  • The appearance of abnormal neurological signs demands the prompt discontinuation of Metronidazole therapy.
  • Metronidazole should be administered with caution to patients with a history of seizures or CNS diseases. 
  • Alcoholic beverages should be avoided during treatment.
  • Metronidazole should be used with caution in patients with blood dyscrasias.
  • Metronidazole should be used with caution in patients with liver impairment, and the dose should be reduced in severe cases.
  • Dosage should be reduced in patients with renal failure (creatinine clearance ≤ 10 ml/min).

    • PRECAUTIONS

  • Metronidazole has been reported to potentiate the anticoagulant effect of Warfarin and other oral anticoagulants.
  • Metronidazole may mask the presence of syphilis.
  • Metronidazole should be avoided during breastfeeding.
  • A dark reddish coloration of urine may occur during the treatment.

    • SIDE EFFECTS

    Side effects are rare and usually mild in nature. These side effects may include gastrointestinal disturbances (e.g. nausea, diarrhea, metallic taste in the mouth), headache, vertigo and skin rashes.

    OVERDOSAGE

    In case of overdose, management should be symptomatic and supportive.

    Store at room temperature (below 25 degrees).
